چکیده

Purpose: To determine the effect of fresh soy milk and its compounds (coumestrol, daidzein genistein) on apoptosis in human leukemic cells peripheral blood mononuclear (PBMC).
Methods: The apoptotic (K562, Jurkat, U937) PBMC was determined by flow cytometry. from healthy donors were isolated conducting density gradient centrifugation principle. Lymphoprep cytotoxicity evaluated 3-(4,5- ‘;1dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) MTT) assay.
Results: treatment genistein had a significantly higher half-maximal inhibitory concentration (IC50) value (p < 0.01) 0.0001), compared to leukemia cells. In addition, IC50 0.05) than Coumestrol induced increased K562, U937 Jurkat 0.05). However, only K562 U937. Genistein reported cell 0.001). Nonetheless, PBMC, significant did not occur. 0.05).
Conclusion: findings indicate that coumestrol, daidzein, inhibits viability induction, but they are non-toxic toward normal Therefore, results suggest compound alone, thus, might have therapeutic leukemia.

Optimization of human peripheral blood mononuclear cells (PBMCs) cryopreservation
Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...

Background & Aims: Verapamil as a calcium channel blocker has been broadly used in the treatment of many cardiovascular diseases such as hypertension and arrhythmia. Furthermore, the anti-tumor/ antiinflammatory effects of verapamil have been shown. In the present study, the cytotoxic effect of verapamil on human peripheral blood mononuclear cells (PBMCs) and BALB/c peritoneal macrophges has be...
متن کاملEffect of Myeloma-Derived Microparticles on in Vitro Proliferation and Viability of Alloimmune Human Peripheral Blood Mononuclear Cells
Background: Microparticles (MPs) are small vesicles released from the cell membrane. Accordingly, these contain active molecules to mediate biological processes, including cell proliferation and cell cycle progression. The fusion of myeloma cell lines with immunized B lymphocytes is a critical step of hybridoma technology. MPs modulate lymphoproliferation, thereby facilitating B cell expansion ...

15 صفحه اولMonoglycerides induce apoptosis in human leukemic cells while sparing normal peripheral blood mononuclear cells.
A major drawback of the current antineoplastic treatments is their lack of specificity toward cancer cells, because they are most often cytotoxic to normal cells, thus creating related side effects. Hence, the identification of new apoptosis-inducing agents, specifically targeting malignant cells while sparing their normal counterparts, is of crucial interest. We show here that monoglycerides, ...
